CY7-SE triethylamine is a CY dye. CY, short for Cyanine, is a compound consisting of two nitrogen atoms connected by an odd number of methyl units. Cyanine compounds have the characteristics of long wavelength, adjustable absorption and emission, high extinction coefficient, good water solubility and relatively simple synthesis. CY dyes are of en used for the labeling of proteins, antibodies and small molecular compounds. For the labeling of protein antibodies, the combination can be completed through a simple mixing reaction. Below, we introduce the labeling method of protein antibody labeling, which has certain reference significance. |

Protocol
一、蛋白制备
1) 为获得最佳标记效果，请制备蛋白 (抗体) 浓度为2 mg/mL。
2) 蛋白溶液的PH值为8.5±0.5。如果pH值低于8.0，应使用1M碳酸氢钠进行调节。
3) 如果蛋白浓度低于2mg/mL，标记效率会大大降低。为获得最佳标记效率，建议终蛋白浓度范围为2-10 mg/mL。
4) 蛋白必须在不含伯胺 (如Tris或甘氨酸) 的缓冲液中，并且铵离子，否则会影响标记效率。
2.染料制备 (以CY3-NHS酯为例)
将无水DMSO加入CY3-NHS酯小瓶中，制成10 mM储备液。通过移液器或涡旋混合均匀。
3.染料用量的计算
反应所需的CY3-NHS酯的量取决于待标记蛋白的量，以及CY3-NHS的最佳摩尔比约为10.
Example：假设所需的marker protein为500 μL 2 mg/mL IgG (MW=150，000)，用100 μL DMSO溶解1 mg CY3-NHS ester，得到所需的CY3-NHS ester体积为5.05 μL，详细计算过程如下：
1) mmol (IgG) =mg/mL (IgG) ×mL (IgG)/MW (IgG) =2 mg/mL × 0.5 mL/150，000 mg/mmol= 6.7×10-6 mmol
2) mmol (CY3-NHS酯) =mmol (IgG) × 10=6.7×10-6 mmol×10=6.7 × 10-5 mmol
3) uL (CY3-NHS 酯) =mmol (CY3-NHS 酯) ×MW (CY3-NHS 酯)/mg/μL (CY3-NHS 酯) =6.7 ×10-5 mmol ×753.88 mg/mmol/0.01 mg/μL=5.05 μL (CY3-NHS 酯)
4.运行偶联反应
1) 将适量新鲜制备的 10 mg/mL CY3-NHS 酯缓慢加入到 0.5 mL 蛋白质样品中
在溶液中，轻轻摇动混匀，然后短暂离心，将样品收集在反应管底部。请勿混匀，以免蛋白样品变性失活。
2) 将反应管置于避光处，在室温下间隔轻轻孵育60分钟。10-15分钟，轻轻逆转反应
5.纯化偶联物
以下方案是使用SepHadex G-25柱纯化染料-蛋白偶联物的示例。
1) 根据制造说明准备SepHadex G-25柱。
2) 将反应混合物 (来自“Run conjugation reaction”) 加载到SepHadex G-25柱的顶部。
3) 一旦样品在顶部树脂表面下方运行，立即添加 PBS (pH 7.2-7.4)。
4) 向所需样品添加更多 PBS (pH 7.2-7.4) 以完成柱纯化。合并含有所需染料-蛋白质偶联物的级分。
