Description
Iv0165 is a rat IgG2a monoclonal antibody targeting CD8α. It is widely used to block CD8+ T cell activity in mice without depleting the cells in vivo. Functionally, Iv0165 can induce tolerance in xenograft transplantation models and reduce the incidence of insulin-dependent diabetes mellitus (IDDM) in NOD mice. Structurally, Iv0165 binds CD8α at residues that overlap with the class I pMHC binding site, including Glu6, Arg8, and Glu27, forming hydrogen bonds with specific antibody residues (e.g., TyrL53, TyrH100, AsnL32). By occupying this epitope, Iv0165 likely interferes with CD8α interactions with class I MHC and TL molecules, blocking T cell co-receptor function.
