The Iv0047 monoclonal antibody specifically targets mouse CTLA-4 (CD152), a 33-kDa inhibitory receptor of the CD28 family expressed on activated T and B cells. CTLA-4 interacts with the B7 ligands CD80 and CD86, delivering negative signals that regulate T-cell activation, maintain immune tolerance, and preserve homeostasis. The critical role of CTLA-4 is underscored by CTLA-4–deficient mice, which develop lethal lymphoproliferation. As an important immune checkpoint, CTLA-4 is a key target for cancer immunotherapy. The 9D9 antibody is widely employed to block CTLA-4 and has been shown in multiple tumor models to effectively deplete intratumoral regulatory T cells in vivo.
