摘要
痤疮是一种常见的慢性炎症性皮肤病,其治疗仍然是一项巨大的临床挑战。在本研究中,我们评估了将多肽 RL-QN15 和 OH-CATH30 结合起来治疗小鼠痤疮的潜力。结果表明,局部应用 RL-QN15 和 OH-CATH30 能显著抑制痤疮丙酸杆菌(P. acnes)的增殖,缓解痤疮引起的水肿。此外,联合治疗还能抑制痤疮丙酸杆菌诱导的促炎细胞因子的过度表达,包括白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α),促进胶原蛋白沉积,从而有效减轻与痤疮相关的皮肤损伤。从机理上讲,RL-QN15和OH-CATH30的组合可抑制小鼠和RAW 264.7细胞中的toll样受体2(TLR2)的表达和核因子卡巴-B(NF-κB)信号通路的激活(P65和IκB的磷酸化)。这些结果表明,这种组合可能会抑制炎症因子的排泄,并通过 TLR2/ NF-κB 信号转导促进胶原蛋白的沉积。总之,我们的研究证明了联合应用 RL-QN15 和 OH-CATH30 的强大治疗效果,强调了 TLR2/NF-κB 通路是痤疮治疗的关键靶点,并为开发创新型痤疮疗法提供了一种新策略。
